Transforming acidic coiled-coil (TACC3), a member of the TACC family, has been shown to be deregulated in various cancers and involved in tumor progression. However, its biological role and molecular mechanism in prostate cancer (PCa) have not been elucidated. Herein, we reported that TACO was markedly upregulated in metastatic PCa. The upregulation of TACO was significantly associated with the metastasis status, tumor stage, total prostate-specific antigen (PSA) level, and Gleason score in patients with PCa. Moreover, a Kaplan-Meier survival analysis showed that patients with PCa who had high TACO expression experienced shorter disease-free survival than patients with a low TACC3 expression. In addition, the knockdown of TACO dramatically reduced the migratory speed and invasiveness of PCa cells. Furthermore, silencing TACC3 markedly suppressed the Wnt/beta-catenin signaling pathway. Taken together, these findings uncover a supportive role for TACC3 in PCa metastasis, which is mediated by the activation of the Wnt/beta-catenin signaling pathway, suggesting that TACO may serve as a prognostic marker in patients with metastatic PCa.
