TTPAL expression as a potential indicator of tumorigenesis and immunosuppression in pancreatic cancer: Evidence from experimental validation and bioinformatic analysis

摘要:
Pancreatic cancer is a highly deadly digestive tract tumor with a high mortality rate and poor prognosis. The challenges in early diagnosis, coupled with the frequent occurrence of drug resistance, underscore the critical need for a deeper understanding of the complex mechanisms driving its pathogenesis. TTPAL is an important gene utilized by RNA sequencing in patients with pancreatic cancer, its notable overexpression in tumor tissue and a strong association with poor patient prognosis identified through Weighted Gene Co-expression Network Analysis (WGCNA), Differential Gene Expression (DEG), and LASSO Cox regression analysis. And TTPAL promotes the proliferation and metastasis of pancreatic cancer cells in vivo and vitro. The more in-depth analysis suggested that the specific immune characteristics, immune cell infiltration, immune checkpoint gene expression, tumor mutational burden (TMB), and microsatellite instability (MSI) are significantly correlated with TTPAL in pancreatic cancer as well as across various other cancer types. Our foundings confirmed that TTPAL is a promising prognostic biomarker for pancreatic cancer and a range of other cancer types. Its oncogenic and immune-regulatory role has been initially validated in the context of pancreatic cancer. Therefore, TTPAL inhibition might offer a promising strategy for pancreatic cancer therapy.Copyright © 2025 The Authors. Published by Elsevier GmbH.. All rights reserved.
